本实验应用 Feulgen 组化染色研究乙胺嘧啶作用后约氏疟原虫卵囊内核酸含量的变化,并在显微分光光度计下进行 DNA 含量测定。结果显示,对照组早、中期卵囊内含有丰富的 DNA 颗粒,晚期卵囊内形成大量成熟的子孢子。而用药组卵囊发育迟缓,胞浆内来见清晰的 DNA 团块,仅见色素堆积,部分卵囊形成黑孢子。用药组卵囊的 DNA 平约含量亦明显低于对照组。初步说明,乙胺嘧啶可抑制疟原虫卵囊的 DNA 合成,从而阻止了蚊体内疟原虫的继续发育成熟.
In this study, Feulgen staining was used to study the changes of nucleic acid in the oocysts of Plasmodium yoelii after pyrimethamine treatment. The content of DNA in the oocysts of Plasmodium yoelii was determined by spectrophotometer. The results showed that the early and mid-term oocysts contained abundant DNA particles in the control group, and a large number of mature sporozoites formed in the late oocysts. The treatment group oocysts stunted cytoplasm to see a clear DNA mass, only pigment accumulation, some of the oocysts form black spores. Drug group oocysts DNA content was also significantly lower than the control group. Preliminary instructions, pyrimethamine can inhibit Plasmodium oocyst DNA synthesis, thereby preventing the mosquito malaria parasite continued to mature.
