目的 :研究大肠癌组织及癌旁正常组织PTPRO基因启动子甲基化状态及m RNA表达情况,并探讨其在大肠癌发生中的关系。方法 :应用甲基化特异性PCR(MSP)和RT-PCR分别检测30例大肠癌组织及癌旁正常组织中PTPRO基因启动子甲基化状态及m RNA表达状态。结果 :大肠癌组织中PTPRO基因启动子甲基化率高于癌旁正常组织,其甲基化样本中PTPRO基因m RNA表达明显受抑。结论 :PTPRO基因启动子甲基化在大肠癌发生中起着重要作用,可能与大肠癌的发生、发展及预后有关。
Objective: To investigate the promoter methylation status and m RNA expression of PTPRO gene in colorectal cancer tissues and adjacent normal tissues, and to explore their relationship in the pathogenesis of colorectal cancer. Methods: Methylation-specific PCR (MSP) and RT-PCR were used to detect the promoter methylation status and m RNA expression of PTPRO gene in 30 cases of colorectal cancer tissues and adjacent normal tissues respectively. Results: The promoter methylation of PTPRO gene in colorectal cancer was higher than that in adjacent normal tissues. The expression of PTPRO m RNA in the methylated samples was significantly inhibited. Conclusion: PTPRO gene promoter methylation plays an important role in the carcinogenesis of colorectal cancer, which may be related to the occurrence, development and prognosis of colorectal cancer.
