本实验观察了胰岛素样生长因子Ⅱ(IGF-Ⅱ)对大鼠离体培养黄体细胞孕酮生成的影响,并对其作用机制进行了探讨。结果显示,IGF-Ⅱ能显著地促进大鼠离体培养黄体细胞孕酮生成并呈剂量-效应关系,同时还能促进3H-亮氨酸(3H-len)掺入黄体细胞蛋白质的合成,促进3H-胸腺嘧啶(3H-TdR)掺入DNA的合成,而上述效应分别被放线菌酮(CYX)和放线菌素D(ActD)所抑制。此外,IGF-Ⅱ对大鼠高体黄体细胞内源性cAMP和hCG诱导的cAMP含量均无影响,这提示IGF-Ⅱ对大鼠黄体细胞孕酮生成的促进作用与其增强黄体细胞蛋白质与DNA的合成有关,而其跨膜信息传递似不通过cAMP系统。
In this study, the effect of insulin-like growth factor Ⅱ (IGF-Ⅱ) on progesterone production in cultured rat luteal cells was investigated and its mechanism of action was also discussed. The results showed that IGF-Ⅱ could significantly promote the progesterone production in rat luteal cells in vitro and in a dose-effect relationship, and at the same time promote the synthesis of 3H-len protein into luteal cells, 3H-thymine (3H-TdR) incorporation into DNA, whereas the above effects are inhibited by cycloheximide (CYX) and actinomycin D (ActD), respectively. In addition, IGF-II had no effect on endogenous cAMP and hCG-induced cAMP levels in rat corpus luteum cells, suggesting that IGF-II may promote progesterone production in rat luteal cells and enhance the protein and DNA of luteal cells Synthesis, and its transmembrane information delivery does not seem to pass cAMP system.
