目的:探讨乳腺癌组织中8号染色体短臂杂合性丢失(LOH)的意义。方法:应用聚合酶链反应技术,检测210例乳腺癌组织中8p上9个及8q上1个座位DNA微卫星多态林记的杂合性丢失。结果:发现在8p11-21.3区域的LOH频率为35%~49%,8p23.1-ter区域的LOH频率为38%~43%,8p22的LOH频率为28%,8q上D8S285的LOH频率仅为24%。结论:研究提示在8p11-21.3及8p23.1-ter区域可能存在与乳腺癌发生发展密切相关的候选抑癌基因。
Objective: To investigate the significance of loss of heterozygosity (LOH) in chromosome 8 of breast cancer. METHODS: Polymerase chain reaction was used to detect the loss of heterozygosity in DNA microsatellite polymorphisms in 9 out of 8 breast cancers and 1 locus on 8 q in 210 breast cancer tissues. RESULTS: It was found that the LOH frequency in the region of 8p11-21.3 was 35% to 49%, the LOH frequency in the region of 8p23.1-ter was 38% to 43%, the LOH frequency of 8p22 was 28%, and the LOH frequency of D8S285 on 8q was only twenty four%. Conclusion: This study suggests that there may be candidate tumor suppressor genes closely related to the development of breast cancer in the 8p11-21.3 and 8p23.1-ter regions.
