目的建立榆树药材中咖啡酸的分析方法。方法以齐墩果酸对照品为对照,采用薄层层析(TLC)对榆树药材进行鉴别;采用高效液相色谱法(HPLC法)测定咖啡酸含量,色谱条件为:色谱柱:Phenomenex Luna C18(2)(4.6mm×150 mm,5μm),流动相:甲醇-磷酸盐缓冲液(23:77),检测波长:323 nm,流速:1.0 ml/min,柱温:35℃。结果采自西藏林芝、山南地区的榆树药材4批,各批榆树药材TLC色谱中均能检出齐墩果酸;咖啡酸HPLC色谱峰与其他色谱峰分离良好,咖啡酸含量在0.039 36~0.236 16μg范围内,呈良好线性关系(r=0.999 8),平均回收率为95.5%,RSD=1.16%。结论 HPLC法简便快捷,结果可靠,为榆树药材的咖啡酸检测提供一个有效方法。
Objective To establish a method for the analysis of caffeic acid in elm trees. Methods Oleanolic acid reference substance was used as a control, TLC was used to identify the elm herbs. The content of caffeic acid was determined by high performance liquid chromatography (HPLC). The chromatographic conditions were: Phenomenex Luna C18 The mobile phase was methanol-phosphate buffer (23:77), the detection wavelength was 323 nm, the flow rate was 1.0 ml / min and the column temperature was 35 ℃ (4.6 mm × 150 mm, 5 μm). Results Four batches of elm medicinal materials from Nyingchi and Shannan of Tibet were obtained. Oleanolic acid was detected by TLC in batches of elm trees. The HPLC chromatograms of caffeic acid were separated from other chromatographic peaks. The content of caffeic acid was 0.039 36 ~ 0.236 16μg range, a good linear relationship (r = 0.999 8), the average recovery was 95.5%, RSD = 1.16%. Conclusion HPLC method is simple, rapid and reliable, providing an effective method for the detection of caffeic acid in elm trees.
