目的:研究RegIV(再生基因Ⅳ)在视网膜母细胞瘤中的表达及其与血管新生的相关性。方法:选择2012年6月-2014年6月在我院行眼球摘除术治疗的视网膜母细胞瘤患儿16例为研究对象。采用免疫组化染色法对16例视网膜母细胞瘤患者肿瘤组织及16例正常视网膜组织标本中的RegIV、VEGF-A(血管内皮生长因子-A)、VEGF-C(血管内皮生长因子-C)的表达进行检测,并分析RegIV的表达与视网膜母细胞瘤患者的临床病理特征及VEGF-A、VEGF-C表达的相关性,同时比较RegⅣ阳性表达病例肿瘤组织与正常视网膜组织中微血管密度(MVD)之间的差异。结果:视网膜母细胞瘤组织中RegⅣ、VEGF-A、VEGF-C的表达水平均较正常视网膜组织显著上升,差异有统计学意义(P<0.05)。视网膜母细胞瘤患者瘤组织中RegⅣ、VEGF-A、VEGF-C表达均与其临床分期、病理分型及视神经侵犯与否均显著相关(P<0.05),与性别、发病部位无关(P>0.05)。视网膜母细胞瘤组织中RegⅣ的表达与VEGF-A表达呈正相关(r=0.696,P<0.05),与VEGF-C表达呈正相关(r=0.728,P<0.05)。视网膜母细胞瘤组织中RegⅣ阳性表达的病例MVD值为(59.67±1.43),明显高于正常视网膜组织中MVD值(30.53±2.31)(P<0.05);视网膜母细胞瘤组织中RegⅣ阴性表达病例MVD值为(38.04±2.86),与正常视网膜组织中MVD值比较无明显差异(P>0.05)。结论:RegIV在视网膜母细胞瘤中呈高表达,且与肿瘤的侵袭与转移相关,其可能通过促进VEGF-A、VEGF-C表达促进血管新生,进而加速视网膜母细胞瘤的发生和进展。
Objective: To investigate the expression of RegIV (regenerative gene Ⅳ) in retinoblastoma and its relationship with angiogenesis. Methods: From June 2012 to June 2014, 16 children with retinoblastoma who were enucleated by enucleation in our hospital were selected as the research object. The expressions of RegIV, VEGF-A (vascular endothelial growth factor-A) and VEGF-C (vascular endothelial growth factor-C) in 16 patients with retinoblastoma and 16 normal retinal tissues were detected by immunohistochemistry The expression of RegIV and the clinicopathological features and the expression of VEGF-A and VEGF-C in patients with retinoblastoma were analyzed. The expression of microvessel density (MVD) in tumor tissues and normal retina tissues was compared between the two groups )difference between. Results: The expression levels of RegⅣ, VEGF-A and VEGF-C in retinoblastoma tissues were significantly higher than those in normal retinas (P <0.05). The expression of RegⅣ, VEGF-A and VEGF-C in patients with retinoblastoma was significantly correlated with their clinical stage, pathological type and optic nerve invasion (P <0.05), but not with sex and disease site (P> 0.05 ). The expression of RegⅣ in retinoblastoma tissues was positively correlated with the expression of VEGF-A (r = 0.696, P <0.05), and positively correlated with the expression of VEGF-C (r = 0.728, P <0.05). The MVD value of RegⅣ-positive cells was (59.67 ± 1.43) in retinoblastoma tissues, which was significantly higher than that in normal retinal tissues (30.53 ± 2.31) (P <0.05). The negative expression of RegⅣ in retinoblastoma tissues The MVD value was (38.04 ± 2.86), which was not significantly different from that in normal retinal tissues (P> 0.05). CONCLUSIONS: RegIV is highly expressed in retinoblastoma and is associated with tumor invasion and metastasis. It may promote angiogenesis by promoting the expression of VEGF-A and VEGF-C, thus accelerating the occurrence and progression of retinoblastoma.
