目的:通过检测环氧化酶-2(COX-2)和Bcl-2蛋白在不同胰腺组织中的表达,探讨COX-2和Bcl-2蛋白在胰腺癌发生发展过程中的作用及其意义.方法:应用免疫组织化学方法PicTureTM通用型二步法检测47例胰腺癌、12例胰腺导管上皮内肿瘤(PIN)和10例正常胰腺组织中COX-2和Bcl-2蛋白的表达.结果:在10例正常胰腺组织中COX-2阳性表达2例,Bcl-2蛋白阳性表达9例;COX-2在PIN和胰腺癌中均呈高表达,分别为75%和68%,明显高于正常胰腺组织(P<0.05).Bcl-2蛋白在PIN组织中阳性表达者占83%,与正常胰腺组织相近(P>0.05),明显高于在胰腺癌组织中47%的阳性表达(P<0.05).胰腺癌组织中COX-2和Bcl-2蛋白的表达呈明显负相关(r=-0.4552,P=0.005).结论:COX-2在PIN和胰腺癌组织中高表达与胰腺癌的发生密切相关,Bcl-2蛋白在胰腺癌发生早期起作用,二者在胰腺癌组织中的表达呈明显负相关.
OBJECTIVE: To investigate the role of COX-2 and Bcl-2 in the development of pancreatic carcinoma by detecting the expression of cyclooxygenase-2 (COX-2) and Bcl-2 protein in different pancreatic tissues. METHODS: The expressions of COX-2 and Bcl-2 protein in 47 cases of pancreatic cancer, 12 cases of pancreatic ductal intraepithelial neoplasia (PIN) and 10 cases of normal pancreas were detected by immunohistochemistry method PicTureTM general two-step method.Results: The positive expression of COX-2 in 10 cases of normal pancreatic tissue was positive in 2 cases and the expression of Bcl-2 was positive in 9 cases. COX-2 was highly expressed in PIN and pancreatic carcinoma, which was 75% and 68% respectively, which was significantly higher than that in normal pancreas (P <0.05) .The positive expression of Bcl-2 protein in PIN tissue was 83%, similar to that in normal pancreatic tissue (P> 0.05), but significantly higher than that in pancreatic carcinoma (47% ). There was a negative correlation between the expression of COX-2 and Bcl-2 protein in pancreatic cancer (r = -0.4552, P = 0.005) .Conclusion: COX-2 overexpression is closely related to pancreatic cancer in PIN and pancreatic cancer Related, Bcl-2 protein in the early stage of pancreatic cancer play a role, both in pancreatic cancer showed a significant negative correlation.
