目的了解先天性白内障小鼠模型与正常小鼠不同发育阶段晶状体蛋白质的差异。方法 ELISA定量检测晶状体蛋白,采用同重同位素相对与绝对定量检测(i TRAQ)。结果随日龄的增长念珠(串珠)状纤维结构蛋白(BFSP1)含量逐渐下降,且白内障小鼠晶状体内的含量低于正常小鼠;随日龄的增长βB2晶状体蛋白含量逐渐上升,且在白内障小鼠晶状体内含量高于正常小鼠。结论不同年龄先天性白内障小鼠与正常小鼠晶状体蛋白质含量差异明显。
Objective To investigate the differences of lens proteins between congenital cataract mice and normal mice at different developmental stages. Methods Quantitative detection of lens protein by ELISA was performed using isotope relative and absolute quantification (i TRAQ). Results With the increase of age, the content of rosary fiber (BFSP1) gradually decreased and the content of lens in cataract mice was lower than that in normal mice. The content of βB2 crystallin increased gradually with age, Mouse lens content higher than normal mice. Conclusion There are significant differences in the content of lens protein in congenital cataract mice and normal mice at different ages.
