目的 :了解短链脂肪酸 ( SCFA)对大鼠移植小肠萎缩及功能低下是否具有预防作用。 方法 :近交系 Wistar大鼠行异位全小肠移植后第 2天开始给予全肠外营养 ( TPN)至第 10天 ,对照组 ( n=10 )行常规 TPN支持 ,SCFA组 ( n=10 )行常规 TPN支持并加用 SCFA,观察移植肠形态学及吸收功能。 结果 :SCFA组移植肠绒毛高度、隐窝深度、粘膜厚度及绒毛面积均明显超过对照组 ,肠上皮细胞偶见线粒体肿大及部分线粒体嵴紊乱、变短变小 ,超微结构明显好于对照组。对照组则出现明显线粒体肿胀 ,嵴短小紊乱和微绒毛萎缩。SCFA组小肠移植大鼠血浆 1 5 N-甘氨酸丰度在 1h、2 h、3h均明显高于对照组。 结论 :短链脂肪酸能维持大鼠移植小肠粘膜形态 ,减轻移植肠上皮细胞超微结构损伤 ,并能改善移植肠对氨基酸的吸收能力
Objective: To investigate whether short chain fatty acids (SCFA) have a preventive effect on atrophy and hypofunction of small intestine in rats. Methods: Total parenteral nutrition (TPN) was given on day 2 after allogeneic small bowel transplantation in allogeneic Wistar rats until day 10, while control group (n = 10) received routine TPN support. SCFA group (n = 10 ) Conventional TPN support and plus SCFA, observed intestinal morphology and absorption function. Results: The intestinal villus height, crypt depth, mucosal thickness and villus area in SCFA group were obviously more than those in control group. The mitochondrial enlargement and some mitochondrial ridges disorder in intestinal epithelial cells were occasionally seen in SCFA group, which became shorter and smaller and the ultrastructure was obviously better than that in control group. The control group showed obvious mitochondrial swelling, short crest disorder and microvilli atrophy. The abundance of plasma 1 5 -glycine in the SCFA group was significantly higher than that of the control group at 1h, 2h, 3h. Conclusion: Short-chain fatty acids can maintain the morphous of small intestine transplanted in rats, reduce the ultrastructural damage of transplanted intestinal epithelial cells and improve the intestinal absorption of amino acids
