将大肠癌TIL在含有rIL-2浓度为60、600和600U/ml的培养基中分别培养扩增,观察不同浓度rIL-2对TIL增殖力、杀伤活性、表型的影响。结果表明:低浓度rIL-2组较中、高浓度组增殖力减弱,但对自身肿瘤细胞的杀伤活性较中、高浓度组增高。三组TIL培养后CD3+细胞比例显著增加,CD4+细胞比例无明显变化,低、中浓度组CD8+细胞比例显著增加,低浓度组CD25+细胞比例显著增加。说明低浓度rIL-2可选择性刺激CD8+、CD25+T细胞克隆增殖,使大肠癌TIL对自身肿瘤杀伤活性提高。
The colorectal cancer TIL was cultured and expanded in culture medium containing rIL-2 at concentrations of 60, 600, and 600 U/ml. The effects of different concentrations of rIL-2 on TIL proliferation, killing activity, and phenotype were observed. The results showed that the proliferative capacity of the low-concentration rIL-2 group was lower than that of the medium and high-concentration groups, but the killing activity against the autologous tumor cells was higher than that of the medium and high concentration groups. After the three groups of TIL cultured, the proportion of CD3+ cells increased significantly, the proportion of CD4+ cells did not change significantly, the proportion of CD8+ cells in the low and medium concentration groups increased significantly, and the proportion of CD25+ cells in the low concentration group increased significantly. It shows that low concentration of rIL-2 can selectively stimulate the proliferation of CD8+ and CD25+ T cells, and enhance the killing activity of colorectal cancer TIL to autologous tumors.
