目的:筛选枸杞多糖抗溃疡的活性部位。方法:采用DEAE-纤维素柱层析法将枸杞多糖分成了4个部分(LBP-1,LBP-2,LBP-3,LBP-4),将昆明种小鼠60只按体重性别均衡随机分为6组:模型组,阳性对照组,LBP-1组,LBP-2组,LBP-3组,LBP-4组,采用无水乙醇(10 mL.kg-1)或消炎痛(20 mg.kg-1)溃疡模型对枸杞多糖抗溃疡有效部位进行筛选,给药剂量为阳性对照组100 mg.kg-1,枸杞多糖组250 mg.kg-1,连续给药7 d,每日2次。结果:LBP-2和LBP-4能够显著抑制无水乙醇造成的小鼠胃溃疡(P<0.05),抑制率分别为77.7%,80.1%,LBP-2也能够显著抑制消炎痛造成的小鼠胃溃疡(P<0.05),抑制率为64.7%。结论:LBP-2和LBP-4为枸杞多糖抗溃疡的活性部位,LBP-2的活性最强。
Objective: To screen the anti-ulcer active site of lycium barbarum polysaccharides. Methods: LBP-1, LBP-2, LBP-3 and LBP-4 were isolated from Lycium barbarum L. by DEAE-cellulose column chromatography. Sixty Kunming mice were randomly divided into three groups The rats in model group, positive control group, LBP-1 group, LBP-2 group, LBP-3 group and LBP-4 group were treated with absolute ethanol (10 mL.kg-1) or indomethacin (20 mg. kg-1) ulcer model of anti-ulcer of LBP were screened, the dose of the positive control group 100 mg.kg-1, LBP 250 mg.kg-1, continuous administration of 7 d, 2 times a day . Results: LBP-2 and LBP-4 significantly inhibited gastric ulcer induced by ethanol (P <0.05), and the inhibitory rates were 77.7% and 80.1%, respectively. LBP-2 also significantly inhibited the mice induced by indomethacin Gastric ulcer (P <0.05), the inhibition rate was 64.7%. CONCLUSION: LBP-2 and LBP-4 are the anti-ulcer active sites of LBP-2, and LBP-2 has the strongest activity.
