目的:探讨大鼠心肌缺血损伤与凋亡蛋白Bcl-2和Bax的关系及牛磺酸的影响。方法:选择健康SD大鼠30只,随机分为假结扎组、结扎组和牛磺酸保护组,每组10只。结扎大鼠冠状动脉左前降支建立心肌缺血模型。检测3组心肌线粒体中超氧化物歧化酶(superoxidedismutase,SOD)、Ca2+-ATP酶活性和丙二醛含量,用免疫组化法检测心肌中的Bcl-2和Bax蛋白。结果:结扎冠状动脉左前降支可致心肌线粒体丙二醛含量升高犤假结扎组:(4.89±0.54)μmol/g;结扎组:(9.85±0.68)μmol/g犦,SOD和Ca2+-ATP酶活性下降犤假结扎组:(8.63±0.35)μmol/(g·s),(13.97±1.97)mmol/(g·s);结扎组:(6.73±0.39)μmol/(g·s),(8.54±2.28)mmol/(g·s)犦。缺血心肌Bax蛋白表达呈显著升高(t=3.931,P<0.01),牛磺酸能明显减少缺血心肌线粒体丙二醛的生成犤牛磺酸保护组:(5.46±0.72)μmol/g犦,降低心肌Bax蛋白的表达,增加Bcl-2蛋白的表达(t=3.716,P<0.01)。结论:结扎大鼠冠状动脉左前降支所致心肌缺血损伤与Bcl-2和Bax蛋白的表达有关,牛磺酸对缺血心肌Bcl-2和Bax蛋白的表达有较好的调控作用。
Objective: To investigate the relationship between myocardial ischemic injury and apoptosis proteins Bcl-2 and Bax and the effect of taurine. Methods: Thirty healthy SD rats were randomly divided into sham group, ligation group and taurine protection group, with 10 rats in each group. Ligation of left anterior descending coronary artery in rats to establish a model of myocardial ischemia. The myocardial mitochondrial superoxide dismutase (SOD), Ca2 + -ATPase activity and malondialdehyde (MDA) content were measured in three groups. The protein levels of Bcl-2 and Bax in myocardium were detected by immunohistochemistry. Results: Ligation of the left anterior descending branch of the coronary artery caused a rise in myocardial mitochondrial malondialdehyde content (4.89 ± 0.54) μmol / g in the ligation group, (9.85 ± 0.68) μmol / g 结, SOD and Ca2 + -ATP (8.63 ± 0.35) μmol / (g · s), (13.97 ± 1.97) mmol / (g · s) in the ligation group: (6.73 ± 0.39) μmol / (g · s) in the ligation group, (8.54 ± 2.28) mmol / (g · s) 犦. The expression of Bax protein in ischemic myocardium was significantly increased (t = 3.931, P <0.01). Taurine significantly decreased the malondialdehyde (MDA) production in ischemic myocardium (6 taurine protection group: (5.46 ± 0.72) μmol / g犦, decreased myocardial Bax protein expression, increased Bcl-2 protein expression (t = 3.716, P <0.01). CONCLUSION: Myocardial ischemia injury induced by ligation of the left anterior descending coronary artery in rats is related to the expression of Bcl-2 and Bax proteins. Taurine may regulate the expression of Bcl-2 and Bax protein in ischemic myocardium.
