目的优选复方虎杖颗粒的提取纯化工艺条件。方法以虎杖苷含量和水浸出物量为综合评价指标,采用L9(34)正交试验考察加水量、煎煮次数、煎煮时间3个因素对提取工艺的影响;以除杂率和虎杖苷转移率为考察指标,采用单因素试验考察醇浓度和清膏相对密度对醇沉工艺的影响。结果复方虎杖颗粒的最佳提取工艺为依次加12,10,10倍量水煎煮3次,每次0.5 h;最佳纯化工艺条件为将提取液浓缩至相对密度1.13(70℃)的清膏,加乙醇至含醇量为80%,静置24 h。结论优化所得提取纯化工艺可最大限度的保留有效成分,并达到了较好的去杂效果,为确定虎杖苷的提取纯化工艺提供了实验依据。
Objective To optimize the extraction and purification of compound Polygonum cuspidatum particles. Methods Polydatin content and water extract volume were used as the comprehensive evaluation indexes. The effects of water addition, boiling times and boiling time on the extraction process were investigated by L9 (34) orthogonal test. The removal rate and polydatin conversion Rate as an indicator, the single factor test to investigate the alcohol concentration and the relative density of clear paste on alcohol precipitation process. Results The optimal extraction process of compound Polygonum cuspidatum granules was followed by decoction of 12, 10, and 10 times amount of water for three times, each time being 0.5 h. The optimum purification conditions were as follows: the extract was concentrated to a relative density of 1.13 (70 ℃) Cream, add ethanol to 80% alcohol content, stand for 24 h. Conclusion The optimized extraction and purification process can retain the maximum effective components, and achieve better removal of impurities, to provide experimental evidence for the extraction and purification of polydatin.
