目的 研究氧化的低密度脂蛋白(oxidized low density lipoprotein OX—LDL)对培养的内皮细胞生长状态的影响及其致凋亡作用。方法用Cu2+引发脂质过氧化过程,制备不同氧化程度LDL,应用不同氧化程度和不同浓度的OX-LDL,分别作用于对数生长期的内皮细胞,用四唑盐比色试验(MTT)检测法检测细胞活性,碘化丙啶(PI)染色法及酶联免疫吸附法(ELISA)检测细胞凋亡。结果MTT细胞活性检测显示,轻度氧化的低密度脂蛋白(mm-LDL)对细胞生长有促进作用。mm—LDL进一步氧化,其促生长作用转换为抑制作用,与对照组比较差异显著(P<0.05)。OX—LDL对细胞生长呈浓度依赖性抑制作用。ELISA法、PI染色法检测凋亡显示,OX—LDL可诱导内皮细胞发生凋亡(与对照组比较P<0.05)。结论 氧化修饰的低密度脂蛋白对内皮细胞的细胞毒性作用与其氧化程度、作用剂量有关,一定浓度的OX—LDL可以导致培养内皮细胞的生长抑制乃至凋亡。
Objective To investigate the effects of oxidized low density lipoprotein OX-LDL on the growth of cultured endothelial cells and its apoptotic effects. Methods Lipid peroxidation induced by Cu2 + was used to prepare LDL with different degrees of oxidation. OX-LDL with different oxidation degree and different concentrations were applied to the endothelial cells in logarithmic growth phase respectively. MTT assay The cell viability was measured by enzyme linked immunosorbent assay (ELISA). The apoptosis of cells was detected by propidium iodide (PI) staining and enzyme - linked immunosorbent assay (ELISA). Results MTT cell activity assay showed that mild oxidized low density lipoprotein (mm-LDL) could promote cell growth. mm-LDL further oxidation, its growth-promoting effect is converted to inhibition, compared with the control group significant difference (P <0.05). OX-LDL inhibited cell growth in a concentration-dependent manner. Detection of apoptosis by ELISA and PI staining showed that OX-LDL could induce endothelial cell apoptosis (P <0.05 compared with control group). Conclusions The cytotoxicity of oxidized low density lipoprotein on endothelial cells is related to the degree of oxidation and the dose of ox-LDL. A certain concentration of OX-LDL can induce the growth inhibition and apoptosis of cultured endothelial cells.
