目的:研究海风藤对人类神经母细胞瘤细胞系列淀粉样前体蛋白(βAPP)基因表达的抑制作用。方法:应用Northern印迹杂交观察βAPPmRNA的变化,应用细胞存活率、乳酸脱氢酶(LDH)释放、细胞集落观察海风藤对神经细胞的直接作用。结果:海风藤选择性的抑制βAPP基因表达。实验浓度内未发现细胞毒性作用。结论:βAPP与Alzheimer’s病(AD)的发病关系密切,海风藤选择性抑制βAPP基因表达为其防治AD提供了一定的依据
OBJECTIVE: To study the inhibition of amyloid precursor protein (βAPP) gene expression in human neuroblastoma cells by Kadurabium. METHODS: Northern blot hybridization was used to observe the change of β-APP mRNA. The cell viability, release of lactate dehydrogenase (LDH), and cell colony were used to observe the direct effect of seaweed on nerve cells. RESULTS: Kadsura selectively inhibited the expression of β-APP gene. No cytotoxic effect was found within the experimental concentration. Conclusion: β-APP is closely related to the incidence of Alzheimer’s disease (AD), and the selective inhibition of βAPP gene expression by seaweed provides a certain basis for the prevention and treatment of AD.
