目的 研究 SMMC- 772 1细胞诱导耐药培养中电镜形态的改变。方法 用流式细胞仪检测 SMMC- 772 1及SMMC- 772 1/ ADM细胞 P-糖蛋白表达 ;用电镜观察 SMMC- 772 1及 SMMC- 772 1/ ADM细胞的超微结构。结果 随着培养液中阿霉素浓度的递增 ,P-糖蛋白表达有逐步增加的趋势 ,培养液中阿霉素达一定浓度时 ,与 SMMC- 772 1细胞相比 SMMC-772 1/ ADM细胞结构发生改变 :多核巨细胞增多 ,内质网增多。结论 随着 P-糖蛋白表达量的增加 ,细胞内质网明显增加 ,说明 SMMC- 772 1细胞形成耐药细胞株 SMMC- 772 1/ ADM后结构形态发生改变
Objective To study the changes of the electron microscopic morphology in SMMC-772 1 cells induced by drug-resistant culture. Methods The expression of P-glycoprotein in SMMC-772 1 and SMMC-772 1 / ADM cells was detected by flow cytometry. The ultrastructure of SMMC-772 1 and SMMC-772 1 / ADM cells was observed by electron microscope. Results As the concentration of doxorubicin in culture medium increased, the expression of P-glycoprotein tended to increase gradually. Compared with SMMC-772 1 cells, the rate of doxorubicin up-regulation in SMMC-772 1 / ADM cells Structural changes: multinucleated giant cells, increased endoplasmic reticulum. Conclusion With the increase of P-glycoprotein expression, the endoplasmic reticulum was significantly increased, indicating that the morphological changes of SMMC-772 1 cells after SMMC-772 1 / ADM cells were formed
