目的:研究皮肤组织在mRNA及蛋白水平表达大麻CB2受体的情况。方法:RTPCR和免疫印迹方法研究CB2受体在大鼠皮肤的表达,免疫组化方法示CB2受体的组织分布。结果:RTPCR方法可检测到大鼠皮肤、脾脏组织中CB2受体在mRNA水平的表达;Western印迹示大鼠皮肤、脾脏组织匀浆中抗CB2抗体,可检测到41.6×103蛋白带,而同为实质脏器的肝脏无CB2受体表达;免疫组化方法证实CB2受体主要分布于皮肤的表皮组织和真皮毛囊组织。结论:CB2受体在皮肤组织mRNA和蛋白水平均有表达,主要分布于表皮和毛囊组织。CB2受体可能是神经系统调节皮肤功能的桥梁。
Objective: To study the expression of CB2 receptor mRNA in skin tissues at mRNA and protein levels. Methods: The expression of CB2 receptor in rat skin was studied by RTPCR and Western blotting. The histological distribution of CB2 receptor was detected by immunohistochemistry. Results: The expression of CB2 receptor in rat skin and spleen was detected by RTPCR method. The anti-CB2 antibody was detected by Western blot in the homogenate of rat skin and spleen, which could detect 41.6 × 103 protein bands, As the real organ of the liver without CB2 receptor expression; immunohistochemistry confirmed that CB2 receptors are mainly distributed in the skin epidermal tissue and dermal hair follicles. Conclusion: CB2 receptors are expressed both in mRNA and protein levels of the skin tissues, mainly in the epidermis and hair follicles. CB2 receptors may be the nervous system regulating the function of the bridge.
