目的探讨桂枝茯苓丸抑瘤作用的分子机制,为桂枝茯苓丸(GFW)开发应用提供实验依据。方法计算抑瘤率,免疫组化法测定PCNA、P21waf/cip蛋白表达,原位杂交法检测Survivin mRNA表达。结果GFW抑瘤率为38.93%,GFW上调肿瘤细胞P21waf/cip表达,下调PCNA及Survivin表达。结论GFW可抑制肿瘤细胞生长,其机制可能与上调P21waf/cip及下调PCNA及Survivin mRNA表达密切相关。
Objective To explore the molecular mechanism of the anti-tumor effect of Guizhi Fuling Pills and provide experimental basis for the development and application of Guizhi Fuling Pill (GFW). Methods The tumor inhibition rate was calculated. The expression of PCNA and P21waf/cip protein was detected by immunohistochemistry and the expression of Survivin mRNA was detected by in situ hybridization. Results The inhibitory rate of GFW was 38.93%. GFW upregulated the expression of P21waf/cip in tumor cells and down-regulated the expression of PCNA and Survivin. Conclusion GFW can inhibit the growth of tumor cells, and its mechanism may be related to the up-regulation of P21waf/cip and down-regulation of PCNA and Survivin mRNA expression.
