目的提取五子衍宗方总多糖并研究其对环磷酰胺(CTX)所致肝损伤的保护作用。方法水提醇沉法提取多糖,Sevag法除蛋白,减压浓缩得到五子衍宗方总多糖。制备CTX致小鼠肝损伤模型,灌胃给予不同剂量五子衍宗方总多糖(375,750 mg/kg),测定肝脏指数,HE染色观察组织形态学改变,检测血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转氨酶(AST)水平和肝组织中超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量。结果与正常对照组相比,模型对照组肝损伤严重,肝细胞广泛变性,肝窦受到挤压,结构不清,肝组织灶状坏死,血清ALT、AST水平升高;MDA含量明显增高,SOD活力显著下降(P<0.05)。与模型对照组比较,五子衍宗方总多糖能明显改善CTX所致的肝损伤程度,降低血清ALT、AST水平;降低MDA含量,增加SOD活力(P<0.05)。结论五子衍宗方总多糖对CTX所致的小鼠肝损伤有保护作用,机制可能与抗氧化应激作用有关。
Objective To extract total polysaccharide from Wuzi Yanzong and study its protective effect against hepatic injury induced by cyclophosphamide (CTX). Methods Water extraction alcohol precipitation method polysaccharide, Sevag method of removing protein, concentrated under vacuum to obtain Wuzi Yanzong Fang polysaccharides. The hepatic injury model induced by CTX was induced in mice. The total polysaccharide (375, 750 mg / kg) was administered intragastrically to different doses of Radix et Rhizoma Radix. The liver index was measured. The histopathological changes were observed by HE staining. Serum alanine aminotransferase (ALT) Aspartate aminotransferase (AST) level and liver tissue superoxide dismutase (SOD) activity, malondialdehyde (MDA) content. Results Compared with the normal control group, the model control group had severe hepatic injury, extensive degeneration of hepatic cells, squamous hepatic sinusoidosis, unclear structure, necrosis of hepatic tissue, elevated serum ALT and AST levels, increased MDA content, Vitality decreased significantly (P <0.05). Compared with the model control group, Wuzi Yanzong Fang polysaccharides could significantly improve the degree of hepatic injury induced by CTX, reduce serum ALT and AST levels, decrease MDA content and increase SOD activity (P <0.05). Conclusion Wuzi Yanzong Fang polysaccharide has a protective effect on CTX-induced liver injury in mice, which may be related to the anti-oxidative stress.
