目的 :探讨山莨菪碱 (6 5 42 )和蜕皮激素 (EDS)对内毒素 (L PS)致伤培养脐静脉内皮细胞(HU VECs)能量代谢的保护作用。方法 :采用反相高效液相色谱分析法测定 L PS损伤后培养 HU VECs ATP、ADP、AMP及能量负荷 (EC)的变化以及 6 5 42、EDS的保护作用。结果 :L PS与内皮细胞孵育后低浓度、早期内皮细胞被激活 ,L PS同一浓度刺激后 ,随刺激时间延长 ,HUVECs EC降低 ,与正常比较 ,12小时后明显降低(P<0 .0 1) ;而用 6 5 42和 EDS治疗后 ,EC、ATP、ADP明显增加 ,其中 EDS治疗后各指标接近正常。结论 :L PS在无血清状态下刺激培养 HU VECs,能量代谢出现紊乱 ,EC、ATP、ADP显著降低。 EDS治疗后 ,EC、ATP、ADP均明显升高 ,且其效果优于 6 5 42。
Objective: To investigate the protective effect of anisodamine (6 5 42) and ecdysone (EDS) on the energy metabolism of human umbilical vein endothelial cells (HU VECs) injured by endotoxin (LPS). Methods: The changes of ATP, ADP, AMP and energy load (EC) in HU VECs after LPS injury and the protective effect of EDS were detected by reverse-phase high performance liquid chromatography. Results: After LPS was incubated with endothelial cells for a short time, the endothelial cells were activated at early stage. The ECs of HUVECs were decreased with the stimulation of LPS at the same concentration, and decreased significantly after 12 hours (P <0.01) ). However, the levels of EC, ATP and ADP were significantly increased after treatment with 6 5 42 and EDS. The indexes of each group were close to normal after EDS treatment. CONCLUSION: L PS stimulates the culture of HU VECs in serum-free state, with disturbance of energy metabolism and significant decrease of EC, ATP and ADP. EDS treatment, EC, ATP, ADP were significantly higher, and the effect is better than 6542.
