目的建立高效液相色谱法测定阿德福韦酯片中阿德福韦酯含量的方法。方法采用高效液相色谱法测定阿德福韦酯的含量,流动相:乙腈-水(含1%三乙胺,磷酸二氢钾50 mmol/L,磷酸调pH至3.0)(40∶60);检测波长:260 nm;流速:1.0 ml/min;柱温:室温。结果阿德福韦酯浓度在4.04~50.50μg/ml范围内线性关系良好(r=0.9999),平均加样回收率为99.77%,RSD为0.24%。结论本方法操作简便,测定结果准确,重复性良好,可用于阿德福韦酯片中阿德福韦酯的含量测定。
Objective To establish a method for the determination of adefovir dipivoxil in adefovir dipivoxil tablets by high performance liquid chromatography. Methods The content of adefovir dipivoxil was determined by high performance liquid chromatography (HPLC). The mobile phase consisted of acetonitrile-water (containing 50 mmol / L triethylamine and potassium dihydrogen phosphate and adjusted to pH 3.0 with phosphoric acid) ; Detection wavelength: 260 nm; flow rate: 1.0 ml / min; column temperature: room temperature. Results Adefovir dipivoxil showed good linearity (r = 0.9999) in the range of 4.04-50.50μg / ml with an average recovery of 99.77% and a RSD of 0.24%. Conclusion The method is simple, accurate and reproducible. It can be used to determine adefovir dipivoxil in adefovir dipivoxil tablets.
