目的:研究槐定碱对内毒素(LPS)致急性肺损伤(ALI)小鼠肺组织中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量及Toll样受体4(TLR4)表达的影响。方法:采用随机数字法,将60只清洁级昆明种小鼠分成6组:正常组、模型组、预防给药组、槐定碱低剂量组、槐定碱中剂量组、槐定碱高剂量组,每组10只。①正常组、②模型组、④槐定碱低剂量组、⑤槐定碱中剂量组、⑥槐定碱高剂量组:ip生理盐水0.5 mL,连续6 d。③预防给药组:ip 5 mg.kg-1槐定碱,连续6 d。上述②~⑥组均在末次注射生理盐水或药物后1 h ip脂多糖(LPS)9 mg.kg-1造模,造模2 h后④~⑥组分别给槐定碱9,5,2.5 mg.kg-1。6 h后检测肺组织匀浆中SOD活性及MDA的含量,取新鲜肺组织裂解后提取RNA,RT-PCR法检测TLR4 mRNA的表达水平。结果:与正常组(86.22±8.86)U.mg-1相比,LPS模型组(50.58±10.06)U.mg-1肺组织匀浆SOD活性明显下降;与模型组相比,各给药组SOD[预防给药组(64.71±5.74)U.mg-1,高剂量组(56.34±6.88)U.mg-1,中剂量组(75.50±13.01)U.mg-1,低剂量组(67.02±10.19)U.mg-1]均明显升高(P<0.05);而肺组织中MDA含量变化趋势与SOD相反(P<0.05);TLR4 mRNA的表达灰度值与正常组(0.45±0.20)比较,LPS模型组(0.82±0.20)明显升高;与模型组比较,各用药组[预防给药组(0.60±0.32)s,高剂量(0.55±0.19),中剂量(0.54±0.29),低剂量组(0.61±0.26)]均明显降低(P<0.05)。结论:槐定碱能降低ALI模型小鼠肺组织匀浆中MDA的活性,提高SOD含量,降低TLR4 mRNA的表达。
AIM: To investigate the effects of sophoridine on the activity of superoxide dismutase (SOD), the content of malondialdehyde (MDA) and the expression of Toll-like receptor 4 (TLR4) in the lungs of mice with acute lung injury induced by endotoxin (LPS) Impact. The above ② ~ ⑥ group were injected intraperitoneal injection of saline or drugs 1 h ip lipopolysaccharide (LPS) 9 mg.kg-1 modeling 2 h after modeling ④ ~ ⑥ group sophoridine 9,5,2.5 mg.kg-1.6 h after the test, the activity of SOD and the content of MDA in the lung homogenate were detected. RNA was extracted from fresh lung tissue and the expression of TLR4 mRNA was detected by RT-PCR. Results: Compared with the normal group (86.22 ± 8.86) U.mg-1, SOD activity in lung homogenate of LPS model group (50.58 ± 10.06) U.mg-1 decreased significantly. Compared with model group, (64.71 ± 5.74) U.mg-1, high dose group (56.34 ± 6.88) U.mg-1, middle dose group (75.50 ± 13.01) U.mg-1 and low dose group ± 10.19) U.mg-1] were significantly higher than those in normal group (P <0.05), while the content of MDA in lung tissue was opposite to that of SOD (P <0.05) (0.60 ± 0.32) s, high dose (0.55 ± 0.19) and middle dose (0.54 ± 0.29) in the LPS model group (0.82 ± 0.20) , Low dose group (0.61 ± 0.26)] were significantly lower (P <0.05). Conclusion: Sophoridine can reduce the activity of MDA in lung homogenate of mice with ALI, increase the content of SOD and decrease the expression of TLR4 mRNA.
