目的研究BMI-l基因在儿童急性白血病患者中的表达及临床意义。方法收集2008年7月1日-2009年4月30日郑州大学第三附属医院和郑州市其他医院的初诊急性白血病患儿标本46例,对照组标本30例。实验经患儿监护人知情同意,并经医院伦理委员会批准。运用反转录-聚合酶链反应(RT-PCR)检测其外周血BMI-1 mRNA表达水平,应用SPSS12.0软件进行数据分析。结果1.BMI-l基因在儿童急性白血病中的表达水平显著高于对照组(P<0.05);2.粒细胞性白血病中该基因部分表达或弱表达,而各型B淋巴细胞性白血病中均有表达,且高于粒细胞性白血病(P<0.05);3.联合化疗未缓解组其表达水平高于缓解组,但二者比较差异无统计学意义(P>0.05),缓解后均未检测到BMI-1 mRNA;4.初治组、复发组与完全缓解组比较,前二者BMI-1 mRNA表达均显著高于后者(P<0.05);初治组、复发组与对照组比较差异有统计学意义(P<0.05),完全缓解组与对照组比较差异无统计学意义(P>0.05)。结论BMI-1基因在儿童急性白血病中表达增高,完全缓解者该基因表达水平趋于正常,提示BMI-1基因可能参与急性白血病的发生、发展过程,并可作为急性白血病的发病、复发及治疗和预后判断的指标之一。
Objective To study the expression of BMI-1 gene in children with acute leukemia and its clinical significance. Methods A total of 46 children with newly diagnosed acute leukemia in the Third Affiliated Hospital of Zhengzhou University and other hospitals in Zhengzhou City were collected from July 1, 2008 to April 30, 2009 and 30 patients in the control group. Experimental informed consent by the child guardian, and approved by the hospital ethics committee. The expression of BMI-1 mRNA in peripheral blood was detected by reverse transcription-polymerase chain reaction (RT-PCR), and SPSS12.0 software was used to analyze the data. The expression of BMI-1 gene in childhood acute leukemia was significantly higher than that in the control group (P <0.05); 2. The gene was partially expressed or weakly expressed in myeloid leukemia, while in all kinds of B lymphocytic leukemia (P <0.05) .3. The expression level in unresponsive group was higher than that in remission group, but there was no significant difference between the two groups (P> 0.05) BMI-1 mRNA was not detected in the newly diagnosed group, relapsed group and complete remission group, the expression of BMI-1 mRNA in the first two groups was significantly higher than the latter (P <0.05) There was significant difference between the two groups (P <0.05). There was no significant difference between the complete remission group and the control group (P> 0.05). Conclusions BMI-1 gene is highly expressed in children with acute leukemia. The expression level of BMI-1 gene is normal in patients with complete remission, suggesting that BMI-1 gene may be involved in the pathogenesis and development of acute leukemia and may be used as the pathogenesis, relapse and treatment of acute leukemia And one of the indicators of prognosis.
