目的:探讨包含有重复序列TTAGGG的寡聚脱氧核糖核苷酸(ODNA151)对不稳定型心绞痛(UAP)患者外周血辅助性T(Th)细胞功能平衡的影响。方法:分离16例UAP患者、10例稳定型心绞痛(SAP)患者和12例胸痛综合征(CPS)患者外周血单个核细胞,培养后应用ELISA法检测Th细胞功能。对UAP患者,在培养体系中加入不同浓度的ODNA151或对照的ODN1612,采用ELISA法测定上清中Th1型细胞因子IFN-γ和Th2型细胞因子IL-4的浓度,评价其对Th亚群功能的影响。结果:与SAP和CPS组比较,UAP组患者Th1细胞功能明显增强。ODNA151抑制UAP患者淋巴细胞培养上清的IFN-γ浓度,并升高IL-4的浓度,降低IFN-γ与IL-4的比值;而对照的ODN1612对细胞因子水平无影响。结论:UAP患者Th1细胞功能明显增强,ODNA151对UAP患者淋巴细胞的Th1/Th2平衡有调节作用,既抑制Th1分化又促进Th2分化。
Objective: To investigate the effect of oligodeoxyribonucleotide (ODNA151) containing repetitive TTAGGG on functional balance of peripheral T helper (Th) cells in patients with unstable angina pectoris (UAP). Methods: Peripheral blood mononuclear cells were isolated from 16 patients with UAP, 10 patients with stable angina pectoris (SAP) and 12 patients with chest pain syndrome (CPS). Th cells were detected by ELISA after culture. To UAP patients, different concentrations of ODNA151 or ODN1612 were added to the culture system, and the concentrations of Th1 cytokines IFN-γ and Th2 cytokines IL-4 in the supernatant were measured by ELISA. Impact. Results: Compared with SAP and CPS groups, the function of Th1 cells in UAP group was significantly enhanced. ODNA151 inhibited the concentration of IFN-γ in lymphocyte culture supernatant of UAP patients, increased the concentration of IL-4, and decreased the ratio of IFN-γ and IL-4. However, ODN1612 did not affect the level of cytokines. Conclusion: The function of Th1 cells in patients with UAP is significantly enhanced. ODNA151 regulates the Th1 / Th2 balance of lymphocytes in patients with UAP, which not only inhibits Th1 differentiation but also promotes Th2 differentiation.
