采用原位杂交和RT-PCR方法,观察消瘀片治疗16周动脉粥样硬化免后对血管壁组织ET-1mRNA和NOSmRNA表达的影响。结果显示,粥样硬化组的ET-1mRNA和iNOSmRNA表达较多,而 cNOSmRNA表达较少。使用消瘀片后,动脉壁组织中的 ET-1mRNA表达明显减少,提示血浆和血管壁组织中的 ET-1减少是由于 ET-1mRNA转录水平降低所致。消瘀片对血管壁组织中iNOSmRNA和cNOSmRNA表达影响是不同的,它可降低iNOSmRNA表达,增加cNOSmRNA表达。表明消瘀片可通过调节 NOS不同亚型基因的表达而消减动脉粥样硬化病变的程度。
In situ hybridization and RT-PCR were used to observe the effect of Xiaoqi Tablets on the expression of ET-1 mRNA and NOS mRNA in the vascular wall tissue after 16 weeks of atherosclerosis-free treatment. The results showed that the atherosclerosis group had more ET-1 mRNA and iNOS mRNA expression, but less cNOS mRNA expression. After the use of Xiaoying Tablets, the expression of ET-1 mRNA in the arterial wall tissue was significantly reduced, suggesting that the decrease in ET-1 in plasma and vascular wall tissue was due to a decrease in the transcription level of ET-1 mRNA. Xiaoying Tablets have different effects on the expression of iNOS mRNA and cNOS mRNA in vascular wall tissue. It can reduce iNOS mRNA expression and increase cNOS mRNA expression. It was shown that Xiaoying Tablets can reduce the degree of atherosclerotic lesions by regulating the expression of different NOS subtype genes.
