目的 :应用傅里叶变换红外光谱法分析大鼠乳鼠的原代培养心肌细胞受高功率微波辐照前后细胞结构的改变情况。方法 :将培养的心肌细胞分为 3组 ,分别用高功率微波辐照 30 ,6 0 ,12 0s,然后在傅里叶变换红外光谱仪的红外显微镜上测定每组细胞的红外光谱。结果与结论 :发现大鼠乳鼠原代培养心肌细胞受高功率微波辐照后 ,细胞膜的结构发生了明显改变。傅里叶变换红外光谱分析技术可用于了解大鼠心肌细胞受微波辐照后细胞膜中蛋白质构象变化 ,尤其是测定细胞膜蛋白质二级结构的变化信息
OBJECTIVE: To study the changes of cell structure of primary cultured cardiomyocytes of rat neonatal rat before and after irradiation with high power microwave by Fourier transform infrared spectroscopy. Methods: The cultured cardiomyocytes were divided into three groups and irradiated with high power microwaves for 30, 60, 120s respectively. Then the infrared spectra of each group of cells were measured by the infrared microscope of Fourier transform infrared spectroscopy. RESULTS AND CONCLUSION: The results showed that the structure of the cell membrane changed significantly after irradiated by high power microwave. Fourier transform infrared spectroscopy can be used to understand the changes of the protein conformation in the cell membrane of rat myocardial cells after microwave irradiation, especially the changes of the secondary structure of the cell membrane proteins
