目的:探讨大肠锯齿状腺瘤和腺癌组织HIF-1α的表达及其与VEGF和血管密度的关系。方法:采用免疫组化法检测HIF-1α和VEGF蛋白的表达。用CD34单克隆抗体标记血管内皮细胞并计数。结果:锯齿状腺瘤HIF-1α蛋白阳性率(62.5%)显著高于正常大肠黏膜组(13.3%),χ2=15.78,P<0.01;显著低于大肠癌组(85.0%),χ2=4.8,P=0.028。锯齿状腺瘤VEGF蛋白阳性率(53.1%)显著高于正常大肠黏膜组(10.0%),χ2=13.17,P<0.01;而与大肠癌组(72.5%)比较差异无统计学意义,χ2=2.89,P=0.089。HIF-1α表达与VEGF正相关(r=0.317,P=0.041);与MVD正相关,r=0.468,P=0.02。结论:HIF-1α促进肿瘤血管生成,参与大肠锯齿状腺瘤癌变。
Objective: To investigate the expression of HIF-1α in the serrated and adenocarcinoma of the large intestine and its relationship with VEGF and vascular density. Methods: Immunohistochemistry was used to detect the expression of HIF-1α and VEGF protein. Vascular endothelial cells were labeled with CD34 monoclonal antibody and counted. Results: The positive rate of HIF-1α protein in serrated adenoma (62.5%) was significantly higher than that in normal colorectal mucosa (13.3%) (χ2 = 15.78, P <0.01), significantly lower than that in colorectal cancer (85.0%) , P = 0.028. The positive rate of VEGF protein in serrated adenoma (53.1%) was significantly higher than that in normal colorectal mucosa (10.0%) (χ2 = 13.17, P <0.01), while it was not significantly different from that in colorectal carcinoma (72.5% 2.89, P = 0.089. HIF-1α expression was positively correlated with VEGF (r = 0.317, P = 0.041), and positively correlated with MVD, r = 0.468, P = 0.02. Conclusion: HIF-1α promotes tumor angiogenesis and is involved in the carcinogenesis of the serrated jejunum of the large intestine.
