目的:研究葛根素在脑缺血早期的脑保护作用是否与脑组织内皮型一氧化氮合酶(eNOS)表达有关。方法:45只大鼠随机分为3个研究组:假手术组(S组)5只、脑缺血组(M组)20只和葛根素预处理组(P组)20只。其中,P组和M组又分别分为脑缺血0.5 h、1 h、2 h、4 h 4个亚组。除假手术组外其它大鼠制作大脑中动脉栓塞(MCAO)模型,P组在手术前10 min腹腔注射葛根素(100 mg/kg),M组和S组在同样时点注射等量生理盐水。采用神经功能缺损评分、神经元尼氏体染色评价脑组织功能及形态变化;采用免疫组化和Western blotting方法检测脑组织eNOS,观察其分布和表达水平。结果:P组各亚组神经功能缺损评分较对应时点的M组减轻(P<0.05),神经元尼氏体溶解也较M组减少,其脑组织eNOS蛋白质的表达均明显高于对应时点的M组(P<0.05)。结论:葛根素预处理对大鼠脑缺血早期的脑保护机制可能与其上调脑组织eNOS蛋白质表达有关。
AIM: To investigate whether the protective effect of puerarin in early stage of cerebral ischemia is related to the expression of endothelial nitric oxide synthase (eNOS) in brain tissue. Methods: Forty five rats were randomly divided into three groups: sham operation group (S group) 5, cerebral ischemia group (M group) 20 and puerarin pretreatment group (P group) 20. Among them, P group and M group were divided into cerebral ischemia 0.5 h, 1 h, 2 h, 4 h 4 subgroups. In addition to the sham operation group, other rats were given MCAO model. Puerarin (100 mg / kg) was intraperitoneally injected in group P at 10 min before operation. M and S groups were injected with the same amount of normal saline . Neurological deficit score and neuron Nissl staining were used to evaluate brain function and morphological changes. Immunohistochemistry and Western blotting were used to detect eNOS in brain tissue and the distribution and expression of eNOS were observed. Results: Neurological deficit score of each subgroup in P group was lower than that in M group (P <0.05), and Nissl lysis of neurons was also decreased compared with M group, the expression of eNOS protein in brain tissue was significantly higher than that of corresponding group Point M group (P <0.05). Conclusion: The mechanism of puerarin preconditioning may be related to the upregulation of brain eNOS protein expression in rats.
