目的研究二苯乙烯苷(TSG)对T2DM大鼠氧化应激、炎症反应及主动脉细胞凋亡相关基因的影响,并探讨其可能机制。方法采用高脂喂养联合低剂量STZ(40mg/kg)的方法建立T2DM大鼠模型后,TSG(60、120mg/kg)、辛伐他汀(SV,2mg/kg)灌胃治疗6周。结果与模型组比较,SV治疗组、TSG治疗组超氧化物歧化酶(SOD)活性、HDL-C水平升高(P<0.05),TG、TC、LDL-C、丙二醛(MDA)、FFA、TNF-α、单核细胞趋化蛋白-1(MCP-1)水平降低(P<0.05)。免疫组化测大鼠主动脉核因子κB(NF-κB)、Fas表达下降(P<0.05),Bcl-2表达增强(P<0.05)。结论 TSG对T2DM大鼠有抗氧化、抗炎作用,并能抑制主动脉细胞凋亡相关基因的表达。
Objective To investigate the effects of stilbene glycoside (TSG) on oxidative stress, inflammatory reaction and aortic apoptosis-related genes in T2DM rats and to explore its possible mechanism. Methods The T2DM rat model was induced by high fat diet combined with low dose of STZ (40mg / kg). TSG (60,120mg / kg) and simvastatin (SV, 2mg / kg) were given intragastrically for 6 weeks. Results Compared with the model group, the activity of SOD, the level of HDL-C and the levels of TG, TC, LDL-C, MDA, The levels of FFA, TNF-α and MCP-1 were decreased (P <0.05). The expression of Bcl-2 and NF-κB in the aorta of rats were decreased (P <0.05), and the expression of Fas was decreased (P <0.05). Conclusion TSG has anti-oxidative and anti-inflammatory effects on T2DM rats and inhibits the expression of apoptosis-related genes in aorta cells.
