目的探讨复方鳖甲软肝片对大鼠单侧输尿管结扎(unilateralureteralobstruction,UUO)肾组织转化生长因子β1(TGFβ1)蛋白及其mRNA表达的影响。方法建立UUO肾间质纤维化模型,随机分为正常对照组、假手术组、模型组、苯那普利组和复方鳖甲软肝片低、中、高剂量等7组。手术后4周处死大鼠,无菌取肾,制作冰冻切片,采用免疫组织化学和原位杂交方法,分别检测肾组织TGFβ1蛋白及其mRNA表达,使用图像分析系统对结果进行处理,采用SPSS11.5统计软件统计分析。结果模型组肾小管上皮细胞和肾间质其他细胞浆内,有少量的TGFβ1蛋白及其mRNA棕黄色颗粒状阳性反应物;苯那普利对照组细胞浆中棕黄色颗粒状阳性反应物明显减少,与模型组比,P<0.01;复方鳖甲软肝片治疗组细胞浆中棕黄色颗粒状阳性反应物明显减少,不同剂量组呈剂量依赖关系,与模型组比,P<0.01。结论TGFβ1是导致肾间质纤维化的重要因素,复方鳖甲软肝片可以通过下调TGFβ1蛋白及其mRNA的表达,对UUO肾间质纤维化大鼠肾脏起一定的保护作用。
Objective To investigate the effect of Fufang Biejia Ruangan Tablet on unilateral ureteral obstruction (UUO) in kidney tissue of transforming growth factor-β1 (TGFβ1) protein and its mRNA expression. Methods UUO renal interstitial fibrosis model was established and randomly divided into normal control group, sham operation group, model group, benazepril group and low dose, medium dose, and high dose of compound Biejia Ruangan Tablets. Four weeks after surgery, the rats were sacrificed and their kidneys were aseptically harvested. Frozen sections were made. Immunohistochemistry and in situ hybridization were used to detect the expression of TGFβ1 protein and its mRNA in the kidney. The results were processed using an image analysis system. SPSS11 was used. 5 statistical software statistical analysis. Results In the tubular epithelial cells of the model group and other cytoplasms of the renal interstitium, there was a small amount of TGFβ1 protein and its mRNA brown-yellow granular positive reaction; in the benazepril control group, the brown-yellow granular positive reaction substance in the cytoplasm was significantly reduced. , Compared with the model group, P<0.01; the positive reaction of brown-yellow granular particles in the cytoplasm of the compound Biejia Ruangan Tablets treatment group was significantly reduced, dose-dependent relationship between different dose groups, compared with the model group, P<0.01. Conclusions TGFβ1 is an important factor leading to renal interstitial fibrosis. Compound Biejia Ruangan Tablets can down-regulate the expression of TGFβ1 protein and its mRNA and play a protective role in kidney of UUO renal interstitial fibrosis rats.
