利用瓶培养富集技术从生产草甘膦工厂排污口的污泥中筛选抗草甘膦菌株,通过菌株形态学鉴定、生理生化特性测定及18S rRNA序列分析,确定其为假丝酵母菌(Candida palmioleophila)。以供试的抗草甘膦菌株总RNA为起始模板,利用Clontech公司的SMARTer技术构建cDNA文库。文库质量鉴定表明:原始文库滴度为2.58×106cfu/mL,扩增后文库滴度为3.42×109cfu/mL,文库库容为2.58×106cfu,重组率为97.6%,插入片段范围为500bp~3kb,主要集中在1kb附近。表明构建的TY-JM的cDNA文库质量符合要求。
Glyphosate-resistant strains were screened from the sludge from the glyphosate factory effluent outlet by bottle culture enrichment technology. The strain was identified as Candida by morphological identification, physiological and biochemical characteristics and 18S rRNA sequence analysis. palmioleophila). The total RNA of the glyphosate-resistant strain tested was used as a starting template, and a cDNA library was constructed using SMARTer technology from Clontech. The library quality identification showed that the titer of the library was 2.58 × 106cfu / mL, the amplified library titer was 3.42 × 109cfu / mL, the library capacity was 2.58 × 106cfu, the recombination rate was 97.6%, and the inserted fragment ranged from 500bp to 3kb. Mainly concentrated in the vicinity of 1kb. The results showed that the quality of the constructed TY-JM cDNA library met the requirement.
