目的:研究慢性束缚应激时大鼠海马与杏仁体BDNF的变化,以及逍遥散对其影响。方法:以慢性束缚应激塑造肝郁证候模型,运用免疫组织化学、Western blot分析及RT-PCR等方法,从蛋白和基因水平,定位与定量相结合,检测BDNF表达水平。结果:在慢性束缚应激条件下,BDNF的表达发生变化,海马和杏仁核往往结果相反。结论:可能是它们由不同的机制控制所引起,而逍遥散对其有一定程度的逆转作用。
Objective: To study the changes of BDNF in hippocampus and amygdala after chronic restraint stress and the effect of Xiaoyaosan on it. Methods: The model of liver depression syndrome was established by chronic restraint stress. The expression of BDNF was detected by immunohistochemistry, Western blot analysis and RT-PCR from protein and gene level, localization and quantification. Results: Under chronic restraint stress, the expression of BDNF changed. The hippocampus and amygdala often had the opposite result. Conclusion: It is possible that they are caused by different mechanisms of control, and Xiaoyao San has a certain degree of reversal.
