目的:建立高效液相色谱法(HPLC法)测定昆明山海棠搽剂体外透皮吸收方法并研究制剂中月桂氮花卓酮的最佳用量。方法:以HPLC法测定雷公藤内酯醇(以下简称甲素)含量。结果:回归方程为Y=0.3265X+0.0266(r=0.9986,n=5)。日内RSD为0.5%(n=5);日间RSD为1.2%(n=5);方法的平均回收率为(92~98)%,RSD≤1.4%(n=5)。月桂氮花卓酮的最佳用量为2.0%,稳态透皮速率J=0.46μg·cm-1·h-1。结论:HPLC法简单易行,可用于甲素含量测定和透皮吸收研究。
OBJECTIVE: To establish a HPLC method for the determination of the transdermal absorption of Kunming Begonia liniment in vitro and to study the optimal dosage of Azone. Methods: HPLC method for the determination of triptolide (hereinafter referred to as A) content. Results: The regression equation was Y = 0.3265X + 0.0266 (r = 0.9986, n = 5). The intra-day RSD was 0.5% (n = 5) and the intraday RSD was 1.2% (n = 5). The average recovery was 92-98% with RSD ≤ 1.4% (n = 5). The optimum dosage of laurocapram was 2.0%, steady state transdermal rate J = 0.46μg · cm-1 · h-1. Conclusion: HPLC method is simple and can be used for the determination of a-formin and transdermal absorption.
