目的 观察内皮素 - 1(ET- 1)促进心钠素 (ANP)分泌作用及内皮素受体拮抗剂 Bosentan对心钠素分泌的影响。方法 通过 SD乳鼠心肌细胞培养 ,分别在无或有 ET- 1(10 - 1 0 mol/L)作用的情况下 ,在细胞培养液中加入 Bosentan(10 - 8m ol/L)干预 ,72 h后收集细胞培养液测定 ANP。结果 ET- 1刺激使心肌细胞分泌 ANP由 (6 .46± 0 .38) ng/ml增加至 (13.6 0± 1.12 )ng/ml,但应用 Bosentan可以明显降低 ANP的分泌 ,由 (13.6 0± 1.12 ) ng/ml降至 (9.42± 0 .5 8) ng/ml,P<0 .0 5。在无 ET- 1刺激时 Bosentan也可使 ANP由 (6 .46± 0 .38) ng/m l下降至 (5 .0 7± 0 .2 6 ) ng/ml,P<0 .0 5。结论 ET- 1可使 ANP分泌增加 ,但不论有无 ET- 1刺激 ,Bosentan均可使 ANP分泌减少。提示 Bosentan能抑制心肌细胞肥大 ,有益于心肌重构的改善
Objective To observe the effect of endothelin - 1 (ET - 1) on the secretion of atrial natriuretic peptide (ANP) and the effect of endothelin receptor antagonist Bosentan on the secretion of atrial natriuretic peptide. Methods Cardiomyocytes from neonatal SD rats were treated with 10 -8 mol / L Bosentan (10-8 mol / L) in the presence or absence of ET - 1 (10 - 10 mol / L) for 72 h The cell culture fluid was collected to measure ANP. Results The ANP secretion of cardiomyocytes was increased from (6.646 ± 0.38) ng / ml to (13.60 ± 1.12) ng / ml by ET-1 stimulation, but the secretion of ANP was significantly decreased by Bosentan (13.6 ± 1.12) ng / ml to (9.42 ± 0.58) ng / ml, P <0.05. Bosentan also decreased ANP from (6.446 ± 0.38) ng / ml to (0.07 ± 0.26) ng / ml in the absence of ET-1 stimulation, P <0.05. Conclusions ET-1 can increase ANP secretion, but Bosentan can reduce ANP secretion with or without ET-1 stimulation.
