目的抗癌口服液抗肿瘤作用的实验研究为该中药的临床应用及深入研究提供理论及实验依据。方法应用MTT法、集落形成率的测定及血清药理学方法,检测该中药在体外对MGC-803胃癌细胞的抗增殖作用及药物敏感性;本实验还采用动物抗移植性肿瘤-小鼠肉瘤S180实验方法研究抗癌口服液的抗肿瘤作用。结果(1)用MTT法和生长曲线的检测结果表明,该中药在体外对MGC-803胃癌细胞株有明显抑制作用,加药组与对照组相比其生长抑制率有显著性差异(P<0.01),而且这种抑制作用呈现浓度和时间的依赖性。(2)通过集落形成率的测定结果表明,加药组与对照组相比其集落形成率和集落形成抑制率有显著性差异(P<0.01),说明该中药对MGC-803胃癌细胞株有明显的抗增殖作用。(3)血清药理学实验结果表明,灌服抗癌口服液的含药血清组对MGC-803胃癌细胞株生长抑制率达28.91%,与阴性对照组相比有显著性差异(P<0.01)。(4)体内实验结果表明,抗癌口服液对小鼠肉瘤S180的实验抑瘤率在小、中、大剂量下分别达到35.9%、39.5%、41.8%。结论抗癌口服液在体内外有显著的抗肿瘤作用。
The purpose of anti-cancer oral anti-tumor effect of experimental study for the clinical application of traditional Chinese medicine and in-depth study provide theoretical and experimental basis. Methods MTT assay, colony formation assay and serum pharmacology were used to detect the antiproliferative effect and drug sensitivity of this Chinese medicine on MGC-803 gastric cancer cell line in vitro. In this experiment, we used animal anti-tumor-bearing sarcoma S180 Experimental Study of Antitumor Oral Liquid Antitumor. Results (1) MTT assay and growth curve test results showed that the Chinese medicine in vitro MGC-803 gastric cancer cell lines were significantly inhibited, compared with the control group, the drug-addition group had a significant difference in the rate of growth inhibition (P < 0.01), and this inhibition showed a concentration and time-dependent. (2) The result of colony formation rate showed that there was significant difference (P <0.01) between the drug-added group and the control group in the colony formation rate and colony formation inhibition rate, indicating that the Chinese medicine has the effect on the MGC-803 gastric cancer cell line Obvious anti-proliferative effect. (3) Serum pharmacology experiment results showed that the drug-containing serum group of anti-cancer oral liquid drenched MGC-803 gastric cancer cell line growth inhibition rate of 28.91%, compared with the negative control group was significantly different (P <0.01) . (4) The results of in vivo experiments showed that the antitumor activity of anti-cancer oral solution on mouse sarcoma S180 was 35.9%, 39.5% and 41.8% at small, medium and large doses, respectively. Conclusion anti-cancer oral liquid in vivo and in vitro have a significant anti-tumor effect.
