采用SPE-HPLC方法研究8种海参的总皂苷特征图谱,比较不同种海参间皂苷的组成及含量差异,为海参的品种鉴定提供依据。使用Zorbax SB-C18(4.6mm×250mm,5μm)色谱柱,以乙腈-1g/L的三氟乙酸溶液为流动相梯度洗脱,柱温30℃,流速1.0mL/min,检测波长205nm。对不同海参体内皂苷类成分进行HPLC分析,所得图谱特征性强,8种海参的各皂苷成分均得到很好的色谱分离。结果表明不同海参体内的皂苷组成及含量有明显差异,所建立的方法可用于海参的品种鉴定。
Using SPE-HPLC method to study the characteristics of total saponins of eight kinds of sea cucumbers, and to compare the composition and content difference of different saponins among sea cucumbers, and to provide evidence for the identification of varieties of sea cucumbers. The mobile phase consisted of acetonitrile-1 g / L trifluoroacetic acid solution with a mobile phase of Zorbax SB-C18 (4.6 mm × 250 mm, 5 μm). The column temperature was 30 ℃, the flow rate was 1.0 mL / min and the detection wavelength was 205 nm. HPLC analysis of the different components of the sea cucumber saponins, the characteristics of the resulting map strong, 8 kinds of sea cucumber each saponin components were well chromatographic separation. The results showed that different sea cucumber in vivo saponin composition and content were significantly different, the established method can be used for identification of varieties of sea cucumber.
