有效的早期诊断是阻断癌前病变、预防癌变发生的必要条件。本实验建立DMBA(二甲基苯并蒽)诱导的金地鼠颊囊致癌模型,采用P_(53)、增殖细胞核抗原(PCNA)等免疫组化方法及VIDAS图像分析,发现P_(53)表达能敏感、准确地反映口腔癌前病变的粘膜病变程度,并观察到P_(53)阳性细胞在不同程度的上皮异常增生间具有不同的分布形态;实验还发现P_(53)与PCNA从不同检测角度揭示了细胞动力学变化的同一本质,初步证实了“P_(53)功能失活后促进了细胞的异常增殖”这一设想;实验结果还提示控制固定时间、注意观察与计数等因素在实验中的重要性。
