目的:研究栀子苷对拟缺血损伤的脑微血管内皮细胞内皮素受体B(endothelin receptor B,EDNRB)的影响,以揭示其在脑缺血中发挥内皮保护作用的分子机制。方法:采用原代培养大鼠脑微血管内皮细胞,氧糖剥夺法制备拟缺血损伤模型,首先用不同浓度的栀子苷进行干预,筛选出栀子苷的最佳浓度,然后分别采用荧光定量PCR和免疫蛋白印迹法检测各组细胞中EDNRB mRNA及蛋白表达水平。结果:栀子苷在33.2μg/ml浓度时能够显著提高拟缺血损伤脑微血管内皮细胞的活性,同时能够从基因到蛋白水平下调EDNRB的表达。结论:栀子苷对拟缺血损伤脑微血管内皮细胞具有保护作用,其机制可能与下调EDNRB的表达有关,这可能是栀子苷在脑缺血性损伤中发挥抗炎作用的内在机制之一。
Objective: To study the effect of geniposide on endothelin receptor B (EDNRB) in ischemic injury of cerebral ischemia and to elucidate its molecular mechanism of endothelial protection in cerebral ischemia. Methods: The model of ischemic injury was prepared by primary culture of rat brain microvascular endothelial cells and oxygen glucose deprivation method. At first, different concentrations of geniposide were used for intervention, and the optimal concentration of geniposide was screened out. Then the fluorescence quantitative The levels of EDNRB mRNA and protein in each group were detected by PCR and Western blotting. Results: Geniposide at a concentration of 33.2μg / ml could significantly increase the activity of ischemic brain injury cells and down-regulate the expression of EDNRB from gene to protein level. CONCLUSION: Geniposide has a protective effect on ischemic injury of rat brain microvascular endothelial cells, and its mechanism may be related to the down-regulation of EDNRB expression, which may be one of the mechanisms underlying the anti-inflammatory effect of geniposide in cerebral ischemic injury .
