为了诱发大鼠慢性肾衰贫血模型并探讨它与血液系统的关系,以含0.4%腺嘌呤的饲料长期饲喂大鼠,并动态检测大鼠外周血象,定期进行肾脏、骨髓检查。成年大鼠普通饮食时血红蛋白(Hb)和红细胞压积(Hct)分别为140.3±10.2g/L和0.42±0.05(n=60),饲喂食0.4%腺嘌呤的饲料约80天,Hb和Hct分别降为104.3±10.2g/L和0.31±0.03,表现为轻、中度贫血,饲喂上述饲料120天则出现重度贫血,Hb和Hct分别为88.1±16.5g/L和0.28±0.06,而外周血白细胞仅有一过性升高,血小板则无明显变化;骨髓形态学及CFU-E培养显示骨髓未受抑制,其间肾脏呈现慢性炎症性病变,肾功能明显降低,血清红细胞生成素(Epo)含量严重不足。结果提示,长期饲喂低含量腺嘌呤饲料,可诱发大鼠慢性肾衰贫血,而该模型与造血系统无直接关系,是研究Epo低下性贫血的理想模型。
In order to induce anemia model of chronic renal failure in rats and to explore its relationship with the blood system, rats were fed with 0.4% adenine for a long time, and the peripheral blood samples of rats were dynamically detected. Kidney and bone marrow examination was performed regularly. Hb and Hct were 140.3 ± 10.2g / L and 0.42 ± 0.05 (n = 60), respectively, while those fed with 0.4% adenine were about 80 days in the normal diet. The results suggest that long-term low-dose adenine feed can induce anemia of chronic renal failure in rats. However, this model is not directly related to the hematopoietic system and is an ideal model for the study of Epo anemia.
