目的:应用大鼠原位循环肠肝灌流模型,考察戊己丸不同配伍对戊己丸中黄连代表成分小檗碱、巴马汀的吸收的影响。方法:采用L9(34)正交设计得到戊己丸12个配伍方(3个单味药和9个配伍方),经十二指肠给药,于不同时间(0,1,5,10,15,30,60,90,120 min)采集灌流液,LC-MS检测小檗碱、巴马汀的含量,计算其吸收分数,吸收速率常数(Ka)。结果:吴茱萸与小檗碱、巴马汀的吸收分数,吸收速率常数(Ka)之间呈负相关,并得到了促进小檗碱、巴马汀吸收的最优配伍比例黄连-吴茱萸-白芍3∶1∶3。结论:吴茱萸抑制黄连代表成分的吸收,而且黄连、吴茱萸、白芍配伍比例不同,小檗碱、巴马汀的吸收量、吸收速度也有较大差异。
OBJECTIVE: To investigate the effects of different compatibility of pentachlorosulfan on the absorption of berberine and palmatine in berberine hydrochloride in Wuji Pills by using the model of intestinal circulation perfusion in rats. Methods: The orthogonal design of L9 (34) obtained 12 compatibility prescriptions of Wuji Pills (3 single herbs and 9 compatibility prescriptions), and administered duodenum at different time (0, 1, 5, , 15, 30, 60, 90, 120 min). The contents of berberine and palmatine were determined by LC-MS. The absorption fraction and absorption rate constant (Ka) were calculated. Results: There was a negative correlation between absorption fraction and absorption rate constants (Ka) of Evodia rutaecarpa and berberine and palmatine, and the optimum ratio of Coptis chinensis - Evodia rutaecarpa Peony 3: 1: 3. Conclusion: Fructus Evodiae can inhibit the absorption of the representative ingredients of Coptis chinensis, and the compatibility ratio of Coptis chinensis Franch, Fructus Evodiae and Radix Paeoniae Alba is different. The absorption and absorption rate of berberine and palmatine are also different.
