目的通过观察RKI-1447对人卵巢癌CAOV-3细胞生长及凋亡的影响,探讨其在体外的抗肿瘤效应。方法体外培养CAOV-3细胞,以不同浓度的RKI-1447为实验组,并设置对照组。CCK8法测细胞增殖抑制效应,光学显微镜观察药物对细胞形态学的影响,流式细胞术检测CAOV-3细胞的凋亡情况。结果 CAOV-3细胞24 h增殖抑制效应有剂量依赖性,RKI-1447浓度为10μmol/L时其抑制效应与其他浓度相比,差异有统计学意义(P<0.05),并可诱导CAOV-3细胞凋亡。结论 RKI-1447能够抑制CAOV-3细胞的生长,其增殖抑制效应具有剂量依赖性,同时可以诱导细胞凋亡。
Objective To observe the effect of RKI-1447 on the growth and apoptosis of human ovarian cancer cell line CAOV-3 and to investigate its antitumor effect in vitro. Methods CAOV-3 cells were cultured in vitro, with different concentrations of RKI-1447 as experimental group and control group. CCK8 method was used to measure the effect of cell proliferation inhibition. The effect of drugs on cell morphology was observed by light microscopy. The apoptosis of CAOV-3 cells was detected by flow cytometry. Results The proliferation inhibitory effect of CAOV-3 cells in a dose-dependent manner was dose-dependent at 24 h. The inhibitory effect of RKI-1447 at a concentration of 10 μmol / L was statistically significant compared with other concentrations (P <0.05) Apoptosis. Conclusion RKI-1447 can inhibit the growth of CAOV-3 cells in a dose-dependent manner and induce cell apoptosis.
