目的:从生理水平初步鉴定宽叶菘蓝体细胞无性系植株的抗病突变体。方法:以一定浓度的根腐病病原菌粗毒素侵染过的同一无性系菘蓝幼叶为实验材料,采用聚丙烯酰胺凝胶电泳技术分析POD和SOD同工酶的谱系,同时测定其POD、CAT、PPO活性。结果:来源于同一无性系植株的POD、SOD酶谱大致相同但也存在部分特异带,染色深浅也有差异且与外部形态有相关性:即长势不好,生根较少的植株,谱带明显加宽,染色也较深;相反,长势良好,生根较多的植株,谱带较浅。POD、CAT、PPO活性变化趋势与植株长势大致相反。结论:以上几种保护酶的酶谱和活性变化为初步筛选抗性突变体和研究菘蓝的抗病机理提供理论依据。
OBJECTIVE: To initially identify disease-resistant mutants of Clonal leaves of Isatis indigotica plants at physiological level. Methods: The young leaves of the same clonal strain infected with crude toxin of root rot pathogen were used as experimental materials, the POD and SOD isozymes were analyzed by polyacrylamide gel electrophoresis, and their POD, CAT, PPO activity. Results: The POD and SOD enzyme profiles of the same clones derived from the same clones were similar, but there were also some specific bands. The depth of staining was also different and correlated with the external morphology: the plants with poor growth and less rooting had significant bands Wide, deep staining; the contrary, growing well, rooting more plants, the spectrum is shallow. POD, CAT, PPO activity trends and plant growth roughly the opposite. CONCLUSION: The changes of the above enzymes and activities of the protective enzymes provide a theoretical basis for the preliminary screening of resistant mutants and the study on the mechanism of disease resistance of Cyperus.
