目的 :研究索拉菲尼(SO)、8-溴-7-甲氧基白杨素(Br MC)及两者合用对SMMC-7721细胞系肝癌干细胞样细胞凋亡的影响。方法 :不同浓度Br MC、SO及两者合用处理体外培养的SMMC-7721细胞系肝癌干细胞样细胞。流式细胞术测定细胞凋亡。Western blot分析NF-κB(p65)蛋白表达。结果 :SO(2.5μmol/L、5μmol/L、10μmol/L、50μmol/L)与Br MC联合处理SMMC-7721细胞系球形成细胞24h,相比于SO单独用药和Br MC单独用药对照组,凋亡率显著增加,同时,NF-κB(p65)蛋白的表达明显下调。结论 :Br MC增强SO诱导SMMC-7721细胞系肝癌干细胞样细胞凋亡作用,其机制涉及下调NF-κB(p65)蛋白表达。
Objective: To investigate the effects of sorafenib (SO), 8-Br-7-methoxychitosan (Br MC) and their combination on the apoptosis of hepatoma stem cell-like cells in SMMC-7721 cells. Methods: SMMC-7721 cells were treated with Br MC, SO and different concentrations of both in vitro and in vivo. Flow cytometry was used to detect apoptosis. Western blot analysis of NF-κB (p65) protein expression. RESULTS: Splenocytes of SMMC-7721 cells were treated with SO (2.5μmol / L, 5μmol / L, 10μmol / L, 50μmol / L) for 24 hours. Compared with SO alone and Br MC alone, The apoptosis rate was significantly increased, meanwhile, the expression of NF-κB (p65) protein was significantly down-regulated. CONCLUSION: Br MC enhances SO-induced apoptosis of hepatoma stem cell-like cells in SMMC-7721 cells by down-regulating NF-κB (p65) protein expression.
