采用空气干燥,热滴片法分别制备强壮箭虫典型种与内海型的染色体标本,用姬姆萨染液对染色体标本进行染色。在显微摄像仪下对清晰的分裂相拍照,计数,分析。实验结果表明:2种类型的强壮箭虫染色体数目、类型一致,都有9对染色体,其中6对为中部着丝点染色体(M);1对为亚中部着丝点染色体(SM);2对为端着丝粒染色体(T)。其核型公式都为2n=18=12M+2SM+4T。采用BSG法对强壮箭虫典型种与内海型染色体进行C-带显色,结果显示:典型种与内海型各条染色体都有C-带分布,C-带分布多在染色体着丝粒和端粒处,两者基本相同,区别是强壮箭虫典型种第3、4号染色体端粒部位比内海型相应部位各多出一条C-带。强壮箭虫典型种与内海型是不同水温下形成的不同世代的多型种。
Using air-drying, hot droplet method were prepared typical muscular insects and internal sea type of chromosome specimens, with Giemsa stain on the chromosome specimens. Under the microscope camera to clear the split phase photo, counting, analysis. The results showed that there were 9 pairs of chromosomes on the two types of strong arrowworms, including 9 pairs of chromosomes, 6 pairs of which were middle centromere (M), 1 pair of middle centromeric chromosomes (SM), 2 Paired telocentric chromosome (T). The karyotype formulas are 2n = 18 = 12M + 2SM + 4T. The BSG method was used to detect the C-band of the typical species and the inland sea-type chromosomes. The results showed that there were C-band distributions in the typical and inland sea-types. The C-band distribution was more in the centromere and end Grain, the two are basically the same, the difference is that the typical type of strong arrow insect on chromosome 3,4 telomere than the corresponding parts of the sea-type more than a C-band. The typical species of strong arrowworms and the inland sea type are multi-species of different generations formed at different water temperatures.
