目的:研究番荔枝内酯化合物 squamocin诱导白血病 HL-60细胞凋亡作用。方法:采用 MTT法检测番荔枝内酯单体化合物 squamocin对白血病细胞株 HL-60细胞增殖的抑制作用; DNA凝胶电泳检测细胞凋亡; Western blot法检测 bc1-2, bax, p21WAF1,磷酸化 p44/42 MAPK的蛋白水平变化。结果: squamocin处理 HL-60细胞 5天后,细胞生长受到明显的抑制, IC50为 0.17μ g/ml。 DNA凝胶电泳可见典型的 DNA梯形带。 squamocin处理 HL 60细胞后, Western blot检测结果呈现 bc1-2, bax, p21WAF1蛋白表达无明显变化,而磷酸化的 MAPK量显著减少。结论: squamocin能诱导 HL-60细胞凋亡,且与 bc1-2、 bax、 p21WAF1蛋白表达水平无关。 MAPK活性抑制可能对其诱导 HL-60细胞凋亡起一定的作用。
Objective: To study the apoptosis of leukemic HL-60 cells induced by squamocin. METHODS: The inhibitory effect of squamocin, an annonaceous squamosin compound, on the proliferation of leukemia cell line HL-60 cells was detected by MTT assay; cell apoptosis was detected by DNA gel electrophoresis; and bc1-2, bax, and p21WAF1 were detected by Western blot. Changes in protein levels of p44/42 MAPK. RESULTS: After 5 days of squamocin treatment on HL-60 cells, cell growth was significantly inhibited with an IC50 of 0.17 μg/ml. DNA gel electrophoresis shows typical DNA ladders. After squamocin treatment of HL60 cells, Western blot analysis showed that the expression of bcl-2, bax, and p21WAF1 protein had no significant changes, but the amount of phosphorylated MAPK was significantly reduced. CONCLUSIONS: Squamocin can induce apoptosis in HL-60 cells, and is not related to the expression levels of bcl-2, bax, and p21WAF1 proteins. Inhibition of MAPK activity may play a role in inducing apoptosis of HL-60 cells.
