目的:探讨寒热中药的成分对TRP家族中TRPV1通道蛋白功能的影响。方法:以辣椒素为TRPV1通道激动剂,采用共聚焦显微成像法在体外观察原代培养DRG神经元胞内[Ca2+]i的变化。结果:寒性中药的成分大黄素可显著下调TRPV1通道蛋白的功能,热性中药的成分桂皮醛可显著上调TRPV1通道蛋白的功能;寒性成分黄芩甙对TRPV1通道蛋白功能虽未见显著影响,但有下调趋势,热性成分吴茱萸碱对TRPV1通道蛋白功能也未见显著影响,但有上调趋势。结论:寒热性中药成分对TRPV1功能的调节可能与中药的寒热药性相关,这可能是寒热性中药临床上发挥寒热调节作用的分子机制之一。
Objective: To investigate the effect of cold and heat herbs on the function of TRPV1 channel protein in TRP family. Methods: The changes of [Ca2 +] i in primary cultured DRG neurons were observed by confocal microscopy with capsaicin as TRPV1 channel agonist. RESULTS: Emodin could significantly down-regulate the function of TRPV1 channel protein, and cinnamic aldehyde, a component of traditional Chinese medicine, could up-regulate the function of TRPV1 channel protein. Baicalin, a component of cold-cold syndrome, had no significant effect on TRPV1 channel protein function, However, there was a downward trend. The evodiamine, evodiamine, had no significant effect on TRPV1 channel protein function, but there was an upward trend. Conclusion: The regulation of TRPV1 function may be related to the cold and heat resistance of traditional Chinese medicine, which may be one of the molecular mechanisms by which cold and heat treatment of traditional Chinese medicine clinically play a role in the regulation of cold and heat.
