作者将经人α-干扰素处理的SMMC7721肝癌细胞系作为抗原免疫小鼠,通过细胞融合,筛选得到一株具有分泌一定特异性的抗肝癌单克隆抗休HAb23细胞株,经亚类鉴定HAb23为IgG1。HAb23细胞株性质稳定。在体外连续培养3个月均有抗体分泌。将冻存的HAb23细胞复苏,其克隆的阳性率保持100%。对30例肝细胞性肝癌及其它组织进行免疫组化染色,其中25例肝癌呈阳性,阳性率达83.3%,正常肝组织、肝炎、肝硬化组织均为阴性。
In this study, the human interferon-treated SMMC7721 hepatoma cell line was used as antigen to immunize mice, and a monoclonal antibody against HAb23 with anti-hepatocellular carcinoma secreting a certain specificity was screened by cell fusion. The subclass identified HAb23 as IgG1. HAb23 cell line stable. In vitro continuous culture 3 months have antibody secretion. The cryopreserved HAb23 cells were resuscitated and the positive rate of their clones kept 100%. Immunohistochemical staining was performed on 30 cases of hepatocellular carcinoma and other tissues, of which 25 cases were positive for hepatocellular carcinoma, the positive rate was 83.3%. The normal liver tissues, hepatitis and liver cirrhosis tissues were all negative.
