为研究SSR分子标记辅助提纯选育棉花新品种的可行性,本研究利用SSR分子标记对402系×33系杂交获得的F5高代材料进行分子标记辅助提纯选育分析,并通过前期已筛选的亲本具有多态性的4对SSR分子标记对F5群体、常规提纯选育F6(CKF6)群体及分子标记辅助提纯后F6(MASF6)群体进行遗传分析。结果表明:F5群体遗传相似系数在0.28~1.0之间,纯度为47%;CKF6群体遗传相似系数在0.57~1.0之间,纯度为58.75%,而MASF6群体带型完全一致;在农艺性状相同的情况下,利用SSR分子标记筛选的MASF6相对于CKF6在这些位点的纯合度提高了1.7倍。研究结果说明利用分子标记辅助提纯选育棉花是一项有效可行的方法。
In order to study the feasibility of SSR molecular marker-assisted purification of new cotton varieties, SSR markers were used to analyze the molecular marker-assisted purification of F5 accessions obtained from the crossing of 402 lines × 33 lines. Four pairs of SSR markers were used to carry out genetic analysis on F5 population, routine purification and breeding of F6 (CKF6) population and F6 (MASF6) population after molecular marker assisted purification. The results showed that the genetic similarity coefficients of F5 population ranged from 0.28 to 1.0 with a purity of 47%. The genetic similarity coefficients of CKF6 population ranged from 0.57 to 1.0 with a purity of 58.75%, while those of MASF6 population were completely consistent. In the same agronomic traits In the case, the homozygosity of MASF6 screened by SSR markers at these sites relative to CKF6 increased by 1.7-fold. The results showed that the use of molecular marker assisted purification of cotton is an effective and feasible method.
