目的对从Cohn氏组分Ⅰ+Ⅲ(即FⅠ+Ⅲ)中粗提IgG的工艺条件进行分步优化。方法使用正交试验优化FⅠ+Ⅲ沉淀的溶解条件并通过比较辛酸沉淀后上清液的纯度和收率确定最佳的辛酸加入量。结果最优工艺条件组合为:溶解液醋酸-醋酸钠缓冲液的浓度为0.020 mol/L,pH4.5,溶解温度为4℃,溶解倍数为10倍,以及辛酸沉淀时按20 mL/L的量加入辛酸。结论通过工艺条件的优化可得到具有较高收率和纯度的IgG粗品,有助于提高最终制品的质量。
Objective To optimize the conditions for the crude extraction of IgG from Cohn’s Fraction I + III (FⅠ + Ⅲ). Methods The orthogonal experiment was used to optimize the dissolution conditions of FⅠ + Ⅲ precipitation and to determine the optimum amount of caprylic acid by comparing the purity and yield of supernatant after caprylic acid precipitation. Results The optimum conditions were as follows: the concentration of acetate-sodium acetate buffer was 0.020 mol / L, pH4.5, the temperature was 4 ℃, the solubilization ratio was 10 times, and the volume of caprylic acid precipitation was 20 mL / L Add bitterness Conclusion The crude IgG with higher yield and purity can be obtained through the optimization of the process conditions, which helps to improve the quality of the final product.
