目的:总结国内外关于RSKs在肿瘤MAPK信号通路中的调控作用,及其调节细胞分裂、存活和分化作用的研究进展。方法:应用Medline及CNKI期刊全文数据库检索系统,以“RSKs,MAPK”为关键词,检索1990-01-2008-10的相关文献,共检索到英文文献44篇和中文文献3篇。纳入标准:1)RSKs的来源和分子结构特征;2)RSKs与MAPK通路的关系;3)RSKs与细胞生长和增殖的关系;4)RSKs与疾病的关系。根据纳入标准:精选31篇文献,最后纳入25篇文献。结果:P90核糖体S6蛋白激酶(RSK)家族是胞外信号调节激酶(ERK)的底物,是Raf-MEK-ERK级联信号通路下游重要的一级效应分子,通过磷酸化大量的细胞内蛋白来调节细胞分裂、存活和分化等。结论:RSKs可能是抗肿瘤研究的潜在靶点。
OBJECTIVE: To summarize the regulatory roles of RSKs in tumor MAPK signaling pathway at home and abroad, as well as their research progress in the regulation of cell division, survival and differentiation. Methods: Medline and CNKI journal full-text database retrieval system were searched. The relevant literature from 1990-01-2008-10 was retrieved with “RSKs, MAPK ” as keywords. There were 44 English articles and 3 Chinese articles retrieved. Inclusion criteria: 1) the origin and molecular structure of RSKs; 2) the relationship between RSKs and MAPK pathways; 3) the relationship between RSKs and cell growth and proliferation; 4) the relationship between RSKs and disease. According to inclusion criteria: 31 articles were selected and 25 articles were finally included. RESULTS: The P90 ribosomal S6 kinase (RSK) family is a substrate of extracellular signal-regulated kinase (ERK) and an important primary effector downstream of the Raf-MEK-ERK cascade signaling pathway, which phosphorylates a large number of intracellular Protein to regulate cell division, survival and differentiation. Conclusion: RSKs may be potential targets for anti-tumor research.
